Regulation of cathepsins S and L by cystatin F during maturation of dendritic cells.
Transgenic cystatin F is expressed in model mice only during demyelination; in chronic demyelination, remyelinating ability is lost.
cystatin F is an intracellular cysteine peptidase inhibitor with readily regulated expression
CST7 is secreted, but artificial modification of its C-terminus can induce its endocytic targeting.
Cystatin F was not colocalized with cystatin C (show CST3 ELISA Kits) shich suggests distinct functions for these two cysteine protease (show CTSB ELISA Kits) inhibitors in U937 cells.
analysis of reduction-dependent activation of human cystatin F
Data demonstrate the addition of N-linked sugars to an Asn-X-Cys (show DNAJC5 ELISA Kits) motif in cystatin F and suggest that the mannose 6-phosphate sorting machinery is used to divert cystatin F from the secretory pathway and to mediate its uptake from extracellular pools.
Our results validate cystatin F as a useful biomarker of early pathogenesis in experimental models of prion (show PRNP ELISA Kits) disease, and point to unexpected species-specific differences in the transcriptional responses to prion (show PRNP ELISA Kits) infections.
During the active remyelinating phase, both CysF knockdown (CysFKD) and microglial-selective CatC (show CTSC ELISA Kits) overexpression (CatCOE) showed a worsening of the demyelination in Plp (show C3 ELISA Kits)(4e/-) transgenic mice. Conversely, during the chronic demyelinating phase, CatC (show CTSC ELISA Kits) knockdown (CatCKD) ameliorated the demyelination. Our results suggest that the balance between CatC (show CTSC ELISA Kits) and CysF expression controls the demyelination and remyelination process.
The cystatin F expression in the activated microglia is closely associated with the effect of the A2A receptors, which may be related to the neuroinflammatory responses occurring during the pathological process.
an inhibitory activity other than cystatin F quenches CatL (show CTSL1 PLURAL_@6155@) activity in activated macrophages
Expression of cystatin F indicates ongoing demyelination and remyelination; absence of cystatin F expression indicates cessation of remyelination in the demyelinating area.
The embryo's cystatin C (show CST3 ELISA Kits) and F expression functions as a protective mechanism against the maternal proteinase cathepsin S (show CTSS ELISA Kits).
The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions. This gene encodes a glycosylated cysteine protease inhibitor with a putative role in immune regulation through inhibition of a unique target in the hematopoietic system. Expression of the protein has been observed in various human cancer cell lines established from malignant tumors.
