the Grna and Grnb single and double knock out zebrafish lack any obvious morphological, pathological and biochemical phenotypes
GrnA positively modulates hepatic MET expression both in vivo and in vitro.
Results support further examination of PGRN as a potential marker in neurodegenerative diseases, since PGRN is stable in serum and CSF (show CSF2 Antibodies) and can be measured using ELISA kits from several providers
Our data demonstrates an unknown association between PGRN and GD and identifies PGRN as an essential factor for GCase's lysosomal localization. These findings not only provide new insight into the pathogenesis of GD, but may also have implications for diagnosis and alternative targeted therapies for GD.
Studied levels of salivary visfatin (show NAMPT Antibodies), chemerin (show RARRES2 Antibodies), and progranulin and in gingivitis, periodontitis, as well as healthy patients. Salivary progranulin levels were similar in all groups; chemerin (show RARRES2 Antibodies) was detected at higher levels in the periodontitis group compared to the gingivitis and the healthy groups; visfatin (show NAMPT Antibodies) levels were higher in patients with gingivitis and periodontitis compared to those of healthy subjects.
Induction of PGRN expression may play a role in rheumatoid arthritis immune reaction and PGRN levels could be a useful biomarker in RA inflammatory response, but is not related with Breg cell levels.
Results indicate that CNS progranulin levels are regulated differently from peripheral progranulin levels in Alzheimer's dementia, frontotemporal dementia and amyotrophic lateral sclerosis
This study demonstrated that there appears to be a differential pattern of cerebellar atrophy in the major genetic forms of FTD (show FTL Antibodies), being relatively spared in GRN.
Data show that progranulin is upregulated in neuroendocrine tumors (NETs) and islets of the multiple endocrine neoplasia 1 (show MEN1 Antibodies) protein (MEN1 (show MEN1 Antibodies)) mouse as well as in the serum of patients with pancreatic NETs associated with glucagonoma syndrome.
data indicate that the described variants may cause a loss-of-function, albeit to a lesser extent than GRN null mutations, and hence could be considered as low-penetrant risk factors for neurodegenerative diseases
Progranulin is increased in lipodystrophy.
This study demonstrated that the GRN Thr272fs carriers of fronto-temporo-parietal damage.
These findings not only demonstrate that PGRN is a co-chaperone of HSP70 (show HSP70 Antibodies) and plays an important role in GCase (show GBA Antibodies) lysosomal localization, but may also provide new therapeutic interventions for lysosomal storage diseases, in particular GD.
The results suggest that PGRN may affect the differentiation of retinal precursor cells to photoreceptor cells through the HGF receptor signaling pathway.
PGRN is overexpressed in the early stage of endotoxic shock in mice.
Deleting C1qa (show C1QA Antibodies) gene significantly reduces synaptic pruning by Grn(-/-) microglia and mitigates neurodegeneration, behavioral phenotypes, and premature mortality in Grn(-/-) mice; results uncover a previously unrecognized role of progranulin in suppressing aberrant microglia activation during aging.
CXCL9 (show CXCL9 Antibodies) expression is strongly upregulated in PGRN KO mice and its level is correlated with severity of inflammation in a dermatitis model.
Progranulin suppresses titanium particle induced inflammatory osteolysis by targeting TNFalpha (show TNF Antibodies) signaling.
Study concluded that core progranulin-mediated neurodegenerative phenotypes are penetrant on C57Bl/6 and 129/SvImJ mouse strains, but that genetic background modulates progranulin's role in neuroinflammation and hippocampal biology
ADAM15 (show ADAM15 Antibodies) and acrogranin are present on and associated with the surface of guinea pig spermatozoa; besides both proteins may play a role during sperm-egg binding.
Granulins are a family of secreted, glycosylated peptides that are cleaved from a single precursor protein with 7.5 repeats of a highly conserved 12-cysteine granulin/epithelin motif. The 88 kDa precursor protein, progranulin, is also called proepithelin and PC cell-derived growth factor. Cleavage of the signal peptide produces mature granulin which can be further cleaved into a variety of active, 6 kDa peptides. These smaller cleavage products are named granulin A, granulin B, granulin C, etc. Epithelins 1 and 2 are synonymous with granulins A and B, respectively. Both the peptides and intact granulin protein regulate cell growth. However, different members of the granulin protein family may act as inhibitors, stimulators, or have dual actions on cell growth. Granulin family members are important in normal development, wound healing, and tumorigenesis.
