the Grna and Grnb single and double knock out zebrafish lack any obvious morphological, pathological and biochemical phenotypes
GrnA positively modulates hepatic MET expression both in vivo and in vitro.
Progranulin CSF (show CSF2 Antibodies) concentrations and CSF (show CSF2 Antibodies)/serum progranulin ratio were significantly higher in patients with infectious diseases, with disturbed BBB (show ALMS1 Antibodies) function and with elevated CSF (show CSF2 Antibodies) cell count and presence of oligoclonal bands.
We investigated the effect of progranulin (PGRN) expression on the proliferation and senescence of cervical cancer cells
Mutations in granulin are the genetic causes of frontotemporal lobar degeneration.
GRN mutated FTD (show FTL Antibodies) patients were older at death and more likely to present with non-fluent aphasia. They had TDP-43 (show TARDBP Antibodies) pathology.
PGRN-mediated stimulation of the PI3K (show PIK3CA Antibodies)/Akt (show AKT1 Antibodies)-ERK1/2 (show MAPK1/3 Antibodies)/CDK4/6 (show CDK4 Antibodies)/pRb (show RB1 Antibodies) pathway

that progranulin could promote invasiveness of epithelial ovarian cancer cells through an epithelial mesenchymal transition program directly
The study demonstrates increased PGRN expression at local sites of inflammation and association between PGRN levels, disease activity, and functional impairment in patients with Rheumatoid Arthritis.
Data show that exclusive occurrence of pSer81 progranulin (PGRN) isoform in PGRN-antibody-positive patients.
Progranulin overproduction due to Fli-1 (show FLI1 Antibodies) deficiency may contribute to the constitutive activation of SSc (show CYP11A1 Antibodies) dermal fibroblasts by antagonizing the antifibrotic effect of TNF (show TNF Antibodies).
Study concluded that core progranulin-mediated neurodegenerative phenotypes are penetrant on C57Bl/6 and 129/SvImJ mouse strains, but that genetic background modulates progranulin's role in neuroinflammation and hippocampal biology
Results suggest that PGRN is involved in the development and/or maturation of neuronal networks comprising Purkinje cells in the cerebellum, which may be a prerequisite to normal motor function
PGRN is a key regulator of insulin (show INS Antibodies) resistance and PGRN may mediate its effects, at least in part, by inducing ER stress via the PERK (show EIF2AK3 Antibodies)-eIF2alpha (show EIF2A Antibodies) dependent pathway
PGRN may play a critical role in homeostasis of IVD (show IVD Antibodies), and may serve as a potential molecular target for prevention and treatment of disc degenerative diseases.
These data suggest that targeting activation of the alpha7 nAChR (show CHRNA7 Antibodies) pathway may be beneficial in decreasing neuroinflammation and reversing some of the behavioral deficits observed in progranulin-deficient frontotemporal dementia.
Prosaposin (show PSAP Antibodies) facilitates sortilin (show SORT1 Antibodies)-independent lysosomal trafficking of progranulin.
In conclusion, PGRN may be a promising treatment target for both atherosclerosis and inflammation-related osteoporosis.

Granulins are a family of secreted, glycosylated peptides that are cleaved from a single precursor protein with 7.5 repeats of a highly conserved 12-cysteine granulin/epithelin motif. The 88 kDa precursor protein, progranulin, is also called proepithelin and PC cell-derived growth factor. Cleavage of the signal peptide produces mature granulin which can be further cleaved into a variety of active, 6 kDa peptides. These smaller cleavage products are named granulin A, granulin B, granulin C, etc. Epithelins 1 and 2 are synonymous with granulins A and B, respectively. Both the peptides and intact granulin protein regulate cell growth. However, different members of the granulin protein family may act as inhibitors, stimulators, or have dual actions on cell growth. Granulin family members are important in normal development, wound healing, and tumorigenesis.
