These results suggested that the testis-specific (show THEG Proteins) linker histone, H1T, possesses a specific function to produce the chromatin architecture required for proper chromosome regulation, such as homologous recombination.
Imaging analysis indicated that H1T expression affected nucleolar formation. We concluded that H1T plays a role in rDNA transcription, by distinctively targeting rDNA repeats.
Mutation of TE1 (show TESC Proteins) subelement leads to a drop in H1t promoter activity; mutation of TE2 (show ARD1A Proteins) subelement increased promoter activity in two of four cell lines.
H1t is not replaced during spermatogenesis by H1.1 or H1.2 (show HIST1H1C Proteins) expression in H1t knock out mice.
RFX2 (show RFX2 Proteins) is strongly expressed in pachytene spermatocytes, remains high in early round spermatids, and declines only in advance of nuclear condensation. Maximum expression correlates well with the appearance of H1t.
Analysis of DNA-induced secondary structure of the carboxyl-terminal domain of histone H1 (show H1F0 Proteins) subtypes H1(0 (show H1F0 Proteins))(C-H1(0 (show H1F0 Proteins))) and H1t (C-H1t).
it appears that not the order of up-phosphorylation but the number of the phosphate groups is necessary for regulated chromatin decondensation, thus facilitating the substitution of H1t by transition proteins and protamines.
Histones are basic nuclear proteins responsible for nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is intronless and encodes a member of the histone H1 family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6.
