the findings of the present study provided novel insights into the control of IPO8 transcription, and may enhance understanding regarding RUNX2 (show RUNX2 Proteins) regulatory mechanisms in osteoblast differentiation, bone development, and degenerative bone disease.
Our studies also suggest that nuclear entry is important for the prooncogenic activity of eIF4E (show EIF4E Proteins), at least in this context. These findings position nuclear trafficking of eIF4E (show EIF4E Proteins) as a critical step in its regulation and position the importin 8-eIF4E (show EIF4E Proteins) complex as a novel therapeutic target.
importin (show KPNA4 Proteins) alpha8 functions as a cNLS receptor with distinct cargo specificity, and that heterodimerization by importin (show KPNA4 Proteins) alpha8 is a novel regulatory mode of cNLS binding, in addition to the autoinhibitory regulation by the importin beta (show KPNB1 Proteins) binding domain.
Knockdown of IPO8 reduced the amount of nuclear p65 (show GORASP1 Proteins) following TNF (show TNF Proteins) stimulation. IPO8 binding to p65 (show GORASP1 Proteins) is NLS (show ALDH1A2 Proteins) independent.
TGF-beta (show TGFB1 Proteins)-induced and basal state spontaneous nuclear import of Smad4 (show SMAD4 Proteins) require importin 7 (show IPO7 Proteins) and 8.
IPO8 is the most accurate reference gene for clinical lung specimens.
Study provides evidence that Imp8 is required for cytoplasmic miRNA-guided gene silencing and affects nuclear localization of Ago proteins.
Importin 8 plays a critical role in mediating the cytoplasm-to-nucleus transport of mature miRNAs. IPO8-mediated nuclear transport of mature miRNAs is also dependent on the association of IPO8 with the Argonaute 2 (show EIF2C2 Proteins) complex.
The importin-alpha/beta complex and the GTPase Ran mediate nuclear import of proteins with a classical nuclear localization signal. The protein encoded by this gene is a member of a class of approximately 20 potential Ran targets that share a sequence motif related to the Ran-binding site of importin-beta. This protein binds to the nuclear pore complex and, along with RanGTP and RANBP1, inhibits the GAP stimulation of the Ran GTPase. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.
