This study showed that LIMK2 messenger RNA levels was significantly upregulated in subjects with schizophrenia in laminar and cellular samples.
Actin remodelling factor LIMK2 is a key player in the ciliogenesis control network in which YAP (show YAP1 Antibodies)/TAZ (show TAZ Antibodies) and directional vesicle trafficking are integral components.
Results show that in addition to a potential role in promoting metastasis, changes in LIMK1 (show LIMK1 Antibodies) and LIMK2 expression and/or activity might contribute to AR function in prostate cancer via regulation of microtubule cytoskeletal dynamics.
results highlight the exciting possibility of combining specific LIMK2 inhibitors with anticancer drugs in the treatment of multi-drug resistant cancers
LIMK2 expression was reduced in intestinal tumours of cancer-prone mice and in human colorectal cancer cell lines and tumours. LIMK2 expression progressively decreased with advancing stage of colorectal cancer.
Overexpression of LIMK2 is associated with breast cancer growth and invasiveness.
Nf1/LIMK2 interaction and inhibition allows to directly connect neurofibromatosis type I to actin cytoskeleton remodeling.
A novel activity of Rho in promoting a complex between fascin-1 and LIMK1/2 that modulates the interaction of fascin-1 with actin, was identified.
LIMK2 is a key regulator that acts through different substrates to provide functional links between the actin cytoskeleton and spindle dynamics.
LIMK2 positively regulates the level of Aurora A (show AURKA Antibodies), thereby engaging in a positive-feedback loop, promoting Aurora-A (show AURKA Antibodies)-mediated oncogenic pathways.
These data suggest that osteoblasts can increase their mechanosensitivity under continuous mechanical stimulation by reducing the actin stress fiber formation through inhibiting the LIMK2 expression.
LIMK2 expression was reduced in intestinal tumours of cancer-prone mice.
LIMK2 activity is required for keratinocyte migration in the developing eyelid.
Using RNAi against LIMK2 to inhibit the cytoskeleton reorganization can promote the expression of c-fos gene and thus enhance the mechanosensitivity of c-fos gene in osteoblasts.
Results suggest that constitutive activation of the Rho-ROCK-LIM (show PDLIM5 Antibodies) kinase pathway by extracellular production of lysophosphatidic acid by the action of autotaxin (show ENPP2 Antibodies) is required to maintain the large size of lysosomes in visceral endoderm cells.
LIMK-1 (show LIMK1 Antibodies)/2 double knockout mice were more severely impaired in both ADF/cofilin (show CFL1 Antibodies) phosphorylation and excitatory synaptic function in the CA1 (show CA1 Antibodies) region of the hippocampus.
a novel pathway emanating from the TGF-beta type I receptor (show TGFBR1 Antibodies) and leading to regulation of actin assembly, via the kinase LIMK2.
There are approximately 40 known eukaryotic LIM proteins, so named for the LIM domains they contain. LIM domains are highly conserved cysteine-rich structures containing 2 zinc fingers. Although zinc fingers usually function by binding to DNA or RNA, the LIM motif probably mediates protein-protein interactions. LIM kinase-1 and LIM kinase-2 belong to a small subfamily with a unique combination of 2 N-terminal LIM motifs and a C-terminal protein kinase domain. The protein encoded by this gene is phosphorylated and activated by ROCK, a downstream effector of Rho, and the encoded protein, in turn, phosphorylates cofilin, inhibiting its actin-depolymerizing activity. It is thought that this pathway contributes to Rho-induced reorganization of the actin cytoskeleton. At least three transcript variants encoding different isoforms have been found for this gene.
