This structural arrangement suggests that the cooperative assembly of the dimeric Purbeta repressor on the sense strand of the ACTA2 (show ACTA2 Proteins) enhancer is dictated by the association of each subdomain with distinct purine-rich binding sites within the enhancer.
These findings suggest that the highly conserved modular repeats of Purbeta fold to form a core functional domain, which mediates the specific and high affinity binding of the protein to single-stranded DNA.
Overexpression of Purbeta, but not Puralpha (show PURA Proteins), significantly downregulated GnRH1 (show GNRH1 Proteins) mRNA levels in transiently transfected GT1 (show MYL4 Proteins)-7 cells, suggesting that Purbeta acts as a repressor of GnRH1 (show GNRH1 Proteins) gene transcription.
Purbeta is a single-stranded DNA-binding repressor of vascular smooth muscle alpha-actin (show ACTA2 Proteins) gene transcription
analysis of of DNA binding and localized strand separation by Pur alpha (show PURA Proteins) and comparison with Pur beta
Pur alpha (show PURA Proteins) and Pur beta repress smooth muscle alpha-actin (show ACTA2 Proteins) gene transcription by means of DNA strand-selective cis (show CISH Proteins)-element binding and cell type-dependent protein-protein interactions
analysis of the quaternary structure of recombinant mouse Purbeta
These data demonstrate that Pur proteins collaborate with Sp3 to regulate a transcriptional program that enables muscle cells to remodel their phenotype.
structural role for certain core arginine residues in forming a conformationally stable Pur beta protein capable of physical interactions necessary for smooth muscle alpha-actin (show ACTA2 Proteins) gene repression.
Activation of the normally silent SM alpha A gene in reprogrammed adult cardiomyocytes is linked to elevated interaction of serum response factor with Pur-alpha (show PURA Proteins)/Pur-beta.
Suggest dynamic interplay between transcriptional activators Pur-alpha (show PURA Proteins)/Pur-beta and repressors in regulating SMalphaA (show ACTA2 Proteins) gene output during myofibroblast differentiation.
results show that PUR proteins are capable of binding to alpha-MHC (show MYH6 Proteins) mRNA and attenuate its translational efficiency; also show robust expression of PUR proteins in failing hearts where alpha-MHC (show MYH6 Proteins) mRNA levels are suppressed
findings offer insights into mechanism, energetics, and sequence of Purbeta repressor binding to a biologically relevant, contractile phenotype-regulating cis (show CISH Proteins)-element while revealing the thermodynamic confines of Purbeta-mediated effects on DNA structure
This gene product is a sequence-specific, single-stranded DNA-binding protein. It binds preferentially to the single strand of the purine-rich element termed PUR, which is present at origins of replication and in gene flanking regions in a variety of eukaryotes from yeasts through humans. Thus, it is implicated in the control of both DNA replication and transcription. Deletion of this gene has been associated with myelodysplastic syndrome and acute myelogenous leukemia.
