The structure of the adaptor protein dap12 has been identified in the zebrafish genome.
DAP12-based activating pathway is conserved between bony fish and mammals
The different expression of DAP12 compared to TREM2 (show TREM2 Antibodies) represents the first description of such variable expressivity in Nasu-Hakola disease (NHD) microglia.
the microglial membrane adaptor protein DAP12 was required for both nerve injury- and intrathecal CSF1 (show CSF1 Antibodies)-induced upregulation of pain-related microglial genes and the ensuing pain, but not for microglial proliferation
Impaired signaling by the TREM2 (show TREM2 Antibodies)-DAP12 pathway leads to altered immune responses in phagocytosis, cytokine production, and microglial proliferation and survival, thus contributing to disease pathogenesis. Review.
T cells modified to express a KIR (show GEM Antibodies)-CAR and DAP12 exhibit superior antitumor activity compared with standard first- and second-generation CD3zeta (show CD247 Antibodies)-based CARs in a xenograft model of mesothelioma highly resistant to immunotherapy.
This study reveled that TYROBP having a central role in the bipolar disease and schizophrenia manifestation.
isolated PMNs have an increased proportion of both TREM1 (show TREM1 Antibodies) and DAP12 compared to normal healthy control
results support a role of DAP12 in stabilizing TREM2 (show TREM2 Antibodies)-CTF (show NFIA Antibodies), thereby protecting against excessive pro-inflammatory responses.
Data indicate that NKG2 receptor NKG2E (show KLRC3 Antibodies) was capable of associating with CD94 (show KLRD1 Antibodies) and DAP12 but that the complex was retained intracellularly at the endoplasmic reticulum.
Macrophage traits in BRC cells facilitate the metastatic process and DAP12 expression might promote metastatic homing to bone and liver tissues.
DAP12 has a role in trafficking newly synthesized KIR (show GEM Antibodies) to the cell surface. It interacts with an immature KIR2DS isoform likely initiating in the ER. DAP12also impacts KIR2DS surface stability.
significant role of DAP12 in the initial inflammatory response, bone remodeling and regeneration
DAP12-mediated microglial activation following axotomy promotes pro-inflammatory responses, and thereby accelerates nerve injury-induced neuron death
a previously unrecognized role for DAP12 expression in tissue-resident alveolar macrophages in mediating acute noninfectious tissue injury through regulation of neutrophil trafficking.
DAP12 expression in liver grafts regulates donor mDC (show ADAM11 Antibodies) migration to host lymphoid tissue, alloreactive T cell responses and transplant tolerance
Integrin alphav beta3/Dap12 double-deficient developed profound osteoporosis.
Our study identifies the DAP12/IRAK-M (show IRAK3 Antibodies)/IL-10 (show IL10 Antibodies) to be a novel molecular pathway in APCs (show APCS Antibodies) exploited by mycobacterial pathogens, allowing infection a foothold in the lung.
We conclude that DAP12, partly through TREM1 (show TREM1 Antibodies)/3, is involved in renal inflammation during progression of unilateral ureter obstruction.
Siglec-15 (show SIGLEC15 Antibodies) regulates osteoclast development and bone resorption by modulating receptor activator of nuclear factor kappaB ligand (RANKL (show TNFSF11 Antibodies)) signaling in association with DAP12.
Dap12- and FcRgamma (show FCER1G Antibodies)-deficiency exacerbates Granulocyte-Macrophage Colony-Stimulating Factor (show CSF2 Antibodies)-driven monocyte differentiation and production of inflammatory monocyte-derived dendritic cells.
This gene encodes a transmembrane signaling polypeptide which contains an immunoreceptor tyrosine-based activation motif (ITAM) in its cytoplasmic domain. The encoded protein may associate with the killer-cell inhibitory receptor (KIR) family of membrane glycoproteins and may act as an activating signal transduction element. This protein may bind zeta-chain (TCR) associated protein kinase 70kDa (ZAP-70) and spleen tyrosine kinase (SYK) and play a role in signal transduction, bone modeling, brain myelination, and inflammation. Mutations within this gene have been associated with polycystic lipomembranous osteodysplasia with sclerosing leukoencephalopathy (PLOSL), also known as Nasu-Hakola disease. Its putative receptor, triggering receptor expressed on myeloid cells 2 (TREM2), also causes PLOSL. Multiple alternative transcript variants encoding distinct isoforms have been identified for this gene.
