Background A synthetic peptide inhibitor that irreversibly inhibits caspase-3 and related protease/caspase activity and blocks apoptosis. DEVD is the specific recognition sequence found in poly (ADP-ribose) polymerase (PARP). The inhibitor is designed as a methyl ester to facilitate cell permeability. (CAUTION: If the intended use is on purified or recombinant enzymes, esterase should be added to generate free carboxyl groups).

Application Notes We recommend using 1000-5000X dilutions for inhibiting caspase-3 activity in Jurkat cell culture (e.g., Add 1 µL to 1-5 mL of culture medium). The inhibitor can be added t°Culture medium together with apoptosis inducer or 1 hr before inducer is added. The optimal doses may vary for different cells and culture conditions.
