Background Nuclear factor kappa B (NF-kappaB) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-kappaB mediates the expression of a great variety of genes in response to extracellular stimuli. NF-kappaB is associated with IkB proteins in the cell cytoplasm, which inhibit NF-kappaB activity. IkB is phosphorylated by IkB kinase (IKK) complex that contains IKKalpha, IKKbeta, and IKKgamma. A novel molecule in the IKK complex was recently identified and designated IKKepsilon/IKK-i. IKKepsilon is required for the activation of NF-kappaB by PMA and T cell receptors but not by TNFalpha and IL-1. IKKepsilon/IKK-i message is expressed in a variety of tissues and is inducible by TNFalpha, IL-1, and LPS.
Application Notes IKK-epsilon antibody can be used for detection of IKK-epsilon by Western blot at 0.5 to 1 µg/ml. (Optimal dilution should be determined by user.) An 80 kDa band should be detected. It has no cross response to IKK-alpha, IKK-beta, or IKK-gamma. Antibody can also be used for immunohistochemistry and ELISA and might be suited for other applications not tested so far.

Handling Advice Antibody can be stored at 4 °C, stable for one year. As with all antibodies care should be taken to avoid repeated freeze thaw cycles. Antibodies should not be exposed to prolonged high temperatures. During shipment, small volumes of antibody will occasionally become entrapped in the seal of the product vial. For products with volumes of 200 myl or less, we recommend gently tapping the vial on a hard surface or briefly centrifuging the vial in a tabletop centrifuge to dislodge any liquid in the container’s cap.
