- Tryptophan hydroxylase is the rate-limiting enzyme in the synthesis of serotonin (5-hydroxytryptamine,or 5HT). Tryptophan hydroxylase\atalyzes the iopterin-dependent monooxygenation of tryptophan to5-hydroxytryptophan (5HT), which is subsequently decarboxylated to form the neurotransmitter serotonin.
