Additional Information

Characteristics Synonyms: TCRG, TCRD, T-Cell Receptor gamma, T-Cell Receptor delta, T-Cell Receptor gamma delta
Alternative name T Cell Receptor (TCR) gamma/delta
Immunogen Soluble gamma/delta T-cell receptor.
Isotype IgG1  (Matching secondary antibodies)
Clone Immu510
Specificity This clone recognizes all human gamma/delta T cells regardless the variable genes orjunction regions they express as assessed by Flow Immunofluorescence studies onpolyclonal gamma/delta T-cell lines as well as gamma/delta T-cell clones (1-5). It has beencharacterized in reference 1. It recognizes 1. 6 to 8. 9% of peripheral CD3 positivelymphocytes from 7 healthy adult donors. Species: Human. Others not tested.

Application Details

Protocol Double staining is also possible with purified unlabeled antibody form using the followingprotocol: Double labeling protocol using lyophilized unconjugated form with a CD3 (Phycoconjugated). To 100 µl of whole blood, add 10ul of the reconst. purified antibody. Incubate 15 minutes atroom temperature (18-25°C). Add 3ml of PBS/BSA/NaN3. Centrifuge 5 minutes at 1200 rpm, discard supernatant. Add 100 µl of secondary antibody F(ab')2 goat anti-mouse Ig conjugated to FITC at usualdilution in PBS/BSA/NaN3. Incubate 15 minutes at room temperature. Repeat step 2 (washing). Resuspend cells in 100 µl of PBS/BSA/NaN3 containing 1 mg/ml of total mouse Ig (tosaturate eventual free sites of the goat anti-mouse FITC). Incubate 5 minutes at roomtemperature. Without washing, add 20 µl of the CD3 PE. Incubate 15 minutes at room temperature. Repeat step 2 (washing).
Application Notes Flow Cytometry: 2 g / 5x10e5 cells/test or 10 l of the reconstituted purified antibody. Asthis antibody recognizes a rare population, around 3.6% of peripheral blood lymphocytes(4), it is often preferable to use double staining experiments with another T-cell marker(CD2, CD3, CD4, CD8). For protocol see below. Western blot: Recognizes the gamma/delta complex under non-reducing conditions andthe isolated delta chain on reducing conditions. Immunoprecipitation: Precipitates the three isoforms of the gamma/delta T-cell receptor(2). Immunofluorescence: Gives excellent results on frozen Sections (4). Immunohistochemisty on Paraffin Sections not tested. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
Concentration 0.2 mg/ml (prior to lyophilization)
Buffer PBS with 1 mg/ml BSA as stabilizer.Reconstitution: Restore in 1.0 ml distilled water.
Storage Store lyophilized at 2-8°C. After reconstitution store (in aliquots) at -20°C. Avoid repeated freezing and thawing. Shelf life: one year from despatch.
