    Specificity
    This anibody localizes Dsc3 present in: - all living epidermal layers - in glandular duct cells - in basal matrix cells and outer root sheath of hair follicles - basal and suprabasal cell layers of stratified epithelia (e.g. vagina, tongue, esophagus), basal layer of bladder urothelium and complex epithelium of trachea - thymic reticulum cells All simple epithelia are negative. Intercalated disks of myocardium are negative. Polypeptide reacting: 109 kDa and 100 kDa polypeptides (desmocollin 3 splice isoforms at pI 5.2) from human epidermal desmosomes. Most tested cultured cell lines derivated from stratified squamous epithelia or squamous cell carcinoma are positive (e.g A-431, HaCat). Does not react with Bovine and Rodent.

  • Application Notes
    Immunoblotting (Western). Immunohistochemistry on Frozen Sctions. Immunohistochemistry on Paraffin-Embedded Sections after microwave treatment: 1/10 inPBS ( pH 7.4) for 1h at RT. Recommendations: For better resolution on Frozen Sections preincubation (directly afterfixation) with 0.05-0.2 % Triton X100, for 5-10 min, depending on tissue type, isrecommended (Ref.2).
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background
    Desmocollin 3 is a calcium-dependent glycoprotein that is a member of the desmocollin subfamily of the cadherin superfamily. These desmosomal family members, along with the desmogleins, are found primarily in epithelial cells where they constitute the adhesive proteins of the desmosome cell-cell junction and are required for cell adhesion and desmosome formation.
