Description
Choline acetyltransferase (ChAT) catalyzes the synthesis of the neurotransmitteracetylcholine (ACh) from choline and acetyl-CoA in cholinergic neurons. ChAT serves as aspecific marker for cholinergic neurons in both peripheral and central nervous systems. Dysfunction of cholinergic neurons underlies aspects of clinical symptoms found inneurological and psychiatric disorders such as Alzheimer's disease, Down and Rettsyndromes. Choline acetyltransferase recruits choline for the synthesis of acetylcholine atpreganglionic parasympathetic and sympathetic fibres. It is also present atparasympathetic postganglionic fibres.
Specificity
Cholinergic neurons in brain and central nervous system. Species: Human, Rat, Mouse, Cat, Monkey and Bat. Others not tested.
Application Notes
ELISA (1/250-1/1000): In a simple ELISA with 1 µg of antigen coated per well, the 50% ODwas observed at an antibody dilution of 1: 500. Under the same conditions the end pointwas observed at 1/10,000). Immunohistochemistry on Frozen Sections. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
