Description ATF2 (Activating Transcription Factor 2, CREBP, HB16, CREB2, TREB7) is a member of theATF/CREB family of basic region leucine zipper DNA binding proteins that regulatestranscription by binding to a consensus cAMP response element (CRE) in the promoter ofvarious viral and cellular genes. Many of these genes are important in cell growth anddifferentiation, and in stress and immune responses. ATF2 is a nuclear protein that bindsDNA as a dimer and can form dimers with members of the ATF/CREB and Jun/Fos families. It is a stronger activator as a heterodimer with cJun than as a homodimer. Several isoformsof ATF2 arise by differential splicing. The stable native full length ATF2 is transcriptionallyinactive as a result of an inhibitory direct intramolecular interaction of its carboxy terminalDNA binding domain with the amino terminal transactivation domain. Followingdimerization ATF2 becomes a short lived protein that undergoes ubiquitination andproteolysis, seemingly in a protein phosphatase-dependent mechanism. Stimulation of thetranscriptional activity of ATF2 occurs following cellular stress induced by severalgenotoxic agents, inflammatory cytokines, and UV irradiation. This activation requiresphosphorylation of two threonine residues in ATF2 by both JNK/SAP kinase and p38 MAPkinase. ATF2 is abundantly expressed in brain.
Specificity Recognises ATF2 phosphorylated on threonine 69. Cross reacts with mouse [threonine 51]Species: Human.

Application Details

Protocol Western Blotting ProtocolTransfer gel to PDVF or nitrocellulose membranePlace membrane in plastic tray in blocking buffer for one hour with agitationRinse in wash bufferIncubate in wash buffer plus primary antibody for one hourWash 6 X 5 minutes with wash bufferIncubate in wash buffer plus secondary antibody for one hourWash 6X 5 minutes with wash bufferDetect (e. g. ECL, Amersham according to manufacturers instructions)Wash bufferPBS + 0. 1% Tween 20Blocking bufferWash buffer + 5% dried milk powderThe concentration of antibodies used depends on each antibody, the amount of antigenand the detection method used. Generally, dilution is in the range of a few hundred timesdilution to a few thousand times dilution, but usually has to be determined empirically.
Application Notes Western Blot: 1μg/ml. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
Concentration 1.0 mg/ml
Buffer PBS + 0.1% sodium azide
Storage Store the antibody undiluted at 2-8°C for one month or (in aliquots) at -20°C for longer. Avoid repeated freezing and thawing. Shelf life: one year from despatch.
