Immunogen 12 amino acid peptide specific for the carboxy terminus of the insulin regulatable glucose transporter. A cysteine was included at the N-terminus to accommodate protein conjugation.
Specificity Reacts with IRGT in rat heart plasma membranes. 2 major bands are detected at 65 kD and 43 kD. The 43 kD band is 80 % blocked by the peptide at 0.5 µg/ml. The 65 kD band (non specific) is not blocked by peptide at this concentration.

Background The stimulation of glucose uptake by insulin in muscle and adipose tissue requires translocation of the GLUT4 glucose transporter from intracellular storage sites to the cell surface. Activation of phosphatidylinositol-3-OH kinase (PI3K) is required for this trafficking event, but it is not sufficient to produce GLUT4 translocation. Human insulin resistance involves a defect in GLUT4 traffic and targeting leading to accumulation in a dense membrane compartment from which insulin is unable to recruit GLUT4 to the cell surface.

Application Notes Immunoprecipitation. Western blot (has been used at 1/1000 in blotting of rat heart plasma membranes with20-30 g protein/lane. Immunoblotting. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
