Description
|
Caspases play a major role in the transduction of the apoptotic signal and execution ofapoptosis in mammalian cells. Caspase 3 (CPP32) and Caspase 6 (Mch2) are the majoractive caspases in apoptotic cells, and are activated in response to distinctapoptosis-inducing stimuli and in all cell lines analyzed. Both CPP32 and Mch2 are presentin apoptotic cells as multiple active species. Caspase-6 cleaves nuclear mitotic apparatusprotein (NuMA) and mediates the shrinkage and fragmentation of nuclei.

Application Notes
|
Immunoprecipitation (Denaturated): use Protein G and 20 l antibody/mg protein lysate. Immunohistochemistry on paraffin embedded sections: 1: 50-1: 100 in an ABC method,incubation period of 30 minutes at room temperature. Formalin fixed paraffin embeddedtissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH6.0 prior to immunostaining. Western Blot (1: 100). Positive Control: Tonsil. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
