Description P504S has been recently described as prostate cancer-specific gene that encodes a proteininvolved in the beta-oxidation of branched chain fatty acids. Expression of P504S proteinis found in prostatic adenocarcinoma but not in benign prostatic tissue. It stainspremalignant lesions of prostate: high-grade prostatic intraepithelial neoplasia (PIN) andatypical adenomatous hyperplasia. P504S can be used as a positive marker for PIN. The p63 gene is highly expressed in the basal or progenitor layers of many epithelialtissues. P63 shows remarkable structural similarity to p53 gene. Unlike p53, the p63 geneencodes multiple isotypes with remarkable divergent abilities to transactivate p53 reportergenes and induce apoptosis.

Specificity The combination of P504S and p63 (PIN) may be extremely useful for diagnosing prostaticintraepithelial neoplasia (PIN). P504S stains cytoplasm in prostatic adenocarcinomas andatypical hyperplasia and p63 stains nuclei in normal and benign prostate glands. Cellular Localization: CytoplasmicSpecies: Human. Application Details

Application Notes Immunohistochemistry on Formalin-Fixed Paraffin Embedded Sections: Dilute 1/25-1/50 inan ABC method (30 minutes at room temperature). Formalin fixed paraffin embedded tissue sections require high temperature antigenunmasking with 10mM Citrate buffer, pH 6.0 prior to immunostaining. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
Storage Store the antibody (undiluted) at 2-8°C. Shelf life: one year from despatch.
