  • Application Notes
    ELISA: (Direct): To detect hFractalkine by direct ELISA (using 100 μL/well antibody solution) aconcentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonalantibody, in conjunction with compatible secondary reagents, allows the detection of atleast 0.2-0.4 ng/well of recombinant hFractalkine. (Sandwich): To detect hFractalkine by sandwich ELISA (using 100 μL/well antibodysolution) a concentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylatedpolyclonal antibody, in conjunction with Polyclonal Anti-Human Fractalkine (PP1009P) as acapture antibody, allows the detection of at least 0.2-0.4 ng/well of recombinanthFractalkine. Western Blot: To detect hFractalkine by Western Blot analysis this antibody can be used ata concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hFractalkine is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions.
    Other applications not tested.

    Background
    CX3CL1 is a member of the delta chemokine subfamily that contains a unique CX3C cysteine motif near the N-terminal. Unlike other known chemokines, it is a type 1 membrane protein containing a chemokine domain tethered on a long mucin-like stalk. CX3CL1, a leukocyte chemoattractant, is expressed in various tissues including the brain, heart, lung, kidney, skeletal muscle, and testis. The expression is reported to be up-regulated in endothelial cells and microglia by inflammatory signals.
