Description
|
Granulocyte Macrophage Colony Stimulating Factor (also known as GM-CSF,Colony-stimulating factor, CSF, sargramostim and molgramostin) is produced in responseto a number of inflammatory mediators by mesenchymal cells present in the hemopoieticenvironment and at peripheral sites of inflammation. Granulocyte Macrophage-CSF is ableto stimulate the production of neutrophilic granulocytes, macrophages, and mixedgranulocyte-macrophage colonies from bone marrow cells and can stimulate the formationof eosinophil colonies from fetal liver progenitor cells. GM-CSF can also stimulate somefunctional activities in mature granulocytes and macrophages. GM-CSF receptors showsignificant homologies with other receptors for hematopoietic growth factors, includingIL2-beta, IL-3, IL-6, IL-7, EPO and the Prolactin receptors.

Specificity
|
This antiserum has been heated to 56°C for 30 minutes. In ELISA and otherimmunoreactive assays, this antiserum will recognize both native and recombinant humanGM-CSF in cell supernatants and certain body fluids. This antibody is useful forneutralization of human GM-CSF in bioassays. For Neutralization, it is recommended to incubate the sample with a 1: 400 dilution of theantiserum for at least 4 hours before being tested. A control of similarly diluted normalRabbit IgG is recommended. Add. Information: Endotoxin Content: < 10 pg/ml by LAL method.

This antiserum against Human GM-CSF has been tested for use in Neutralizations, ELISA,Radioimmunoassays, Immunohistochemistry and Immunoblotting. Reactivity in otherimmunoassays is unknown. Recommended Dilutions: This product has been assayed by immunoblot using HRPGoat-anti-Rabbit IgG [H&L] and TMB as a substrate. A working dilution of 1: 400 issuggested for this application. This product has been assayed by Immunohistochemistry. A dilution of 1: 400 or greater is suggested for this immunoassay. Either paraffin fixation orcryofixation can be used for Immunohistochemistry. This product has been assayed byELISA against GM-CSF using HRP Conjugated Anti-Rabbit IgG [H&L] (Goat) and ABTS as asubstrate for 30 minutes at room temperature. A working dilution range of 1: 600 to 1: 1,200is suggested for this product. For use in ELISA formats, this antibody is best used as thesecond antibody in combination with a monoclonal antibody as a capture antibody. Thisproduct has been assayed by Radioimmunoassay against antigen. A dilution of 1: 8,000 issuggested for this immunoassay. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
