    Specificity
    This monoclonal 2D11.H5 antibody recognizes a polypeptide which is identified as Insulin, a 51-amino acid polypeptide composed of A and B chains connected through the C-peptide. This protein A purified Mouse monoclonal antibody reacts specifically with Insulin from Human and Swine sources. Cross reactivity with Insulin from Mouse and Rat does not occur. Cross reactivity with Insulin from other sources has not been determined.

  • Application Notes
    This antibody is suitable for ELISA, Immunohistochemistry, Immunoblotting andimmunoprecipitation. Insulin has a cytoplasmic localization. Pancreatic tissue or β-cellsfrom islets of Langerhans can be used as a positive control. Generally a 1: 50 to 1: 200 dilution is suitable for most applications. A 30 to 60 minuteincubation time is usually sufficient. However, depending upon the fixation conditions andthe staining system employed, optimal incubation should be determined by the user. Frozen sections of paraffin embedded tissues are recommended for preparations to use forimmunohistochemistry. No special pretreatment is required. Researchers shoulddetermine the optimal dilution for other specific assays and cell types. ELISA: Results of Mab anti-Insulin antibody were tested against human insulin by ELISA. Each well was coated with 0.1g of conjugate. The starting concentration of antibody in thedilution series was 10 μg/mL. Each point on the Y-axis represents a 3-fold dilution. Themidpoint of the titration curve represents approx-imately 5 ng/mL antibody or a 1: 200,000dilution from the stock concentration. HRP conjugated Gt-a- Mouse IgG H&L and TMBsubstrate were used for detection (see figure 1).

    Background
    Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-peptide basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain acids. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. The main storage site for insulin is the pancreatic islets. Antibodies to insulin are important as β-cell and tumor (insulinoma) markers.Synonyms: INS
