Immunogen This whole rabbit serum was prepared by repeated immunizations with a synthetic peptide, corresponding to a region (aa 21-40) near the N-terminus of mouse angiopoietin 2 protein conjugated to KLH using maleimide.
Sequence N-N-F-R-K-S-V-D-S-T-G-R-R-Q-Y-Q-V-Q-N-G-P Remarks: A residue of cysteine was added to the amino terminal end to facilitate coupling.
Specificity This antiserum is directed against mouse angiopoietin 2 and shows no reactivity with angiopoietin 1 on mouse derived proteins. A 75 kDa band is detected when assayed by Immunoblot. This reagent cross reacts with human angiopoietin 2. Partially cross reactivity is noted with human angiopoietin 1.

Background Angiopoeitin-1 (Ang-1) and Antiopoietin-2 (Ang2) are important for development of the endothelium, by regulating tyrosine phosphorylation of the membrane receptor Tie-2/Tek. Ang-1 binding to Tie-2/Tek causes phosphorylation of the receptor. Ang-2 competes for this binding, and thus blocks receptor phosphorylation. Ang-2 expression occurs at sites of vascular remodelling: dorsal aorta and major aortic branches, ovary, placenta and uterus. Anti-Ang 1 and anti-Ang 2 antibodies recognize a band above 55 kDa. Both antibodies show banding at 75 kDa which resembles the original size, because these are soluble highly glycosylated proteins, which should run higher that the calculated molecular weight of 55 kD.

Application Notes Suitable for Western blotting (1: 500) and other antibody based assays. The reaction of this antiserum directly with cell supernatants may result in highbackground due to reactivity of components in the serum. This can be alleviated by firstimmunoprecipitating the antibody: antigen complex and then detecting the antigen. Thismethod results in very clean and strong signal. Recommended Dilutions: This product was assayed by immunoblot against supernatantsof angiopoietin expressing endothelial cells from human and mouse sources. A singleband of the expected apparent molecular weight was observed at a 1: 500 dilution. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
