| Description | Group I of the Metabotropic Glutamate Receptors includes GRM1 and GRM5, receptors that have been shown to activate phospholipase C. Alternative splice variants of GRM5 have been described, but their full-length nature has not been determined. Inflammation results in the activation of mGluR1 and mGluR5 in dorsal horn neurons of the spinal cord, leading to extracellular signal-regulated kinase (ERK) activation, which is required for nociceptive plasticity and enhanced pain. GRM5 is involved in inflammatory mechanical hyperalgesia and potentially in schizophrenia. GRM5 has been reported to be expressed in various regions of the brain as well as in the spinal cord and dorsal root ganglia. ESTs have been isolated from human brain, eye, testis, and fetal libraries. GLUR5 refers to Ionotropic Glutamate Receptor 5, not to Metabotropic Glutamate Receptor 5 (mGLUR5). Ligand: Glutamate. | Application Notes | Western Blot. Immunohistochemistry with formalin fixed paraffin embedded tissues: 4-13 ug/ml Antigen retrieval: Steam slides in 0.01 M sodium citrate buffer, pH 6.0, at 99-100°C for 20 min. Remove from heat and let stand at room temperature in buffer for 20 min. Rinse in 1x TBS with Tween (TBST) for 1 min. at room temperature. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user. Further Comments: This product was originally produced by MBL International. |
