Additional Information

Alternative name ADAMTS5
Swiss-Prot Q9UNA0
Immunogen Synthetic peptide corresponding to residues C K(636) N G Y Q S D A K G V K T F(649) of human ADAMTS5
Description A disintegrin-like and metalloproteinase thrombospondin type 1 motif 5 (ADAMTS5 or aggrecanase 2) is a member of the ADAMTS family of multidomain proteins that has demonstrated metalloproteinase functions. The ADAMTS5 protein contains a prohormone convertase cleavage site, thrombospondin-1 like domain, a zinc-binding motif, a disintegrin- like domain, cysteine-rich domain, and a propeptide domain. ADAMTS5 expression is found in a wide range of human tissues, including the placenta, bladder, rib cartilage, cervix and joint tissue. ADAMTS5 plays an important role in the cleavage of aggrecan and studies have determined that aggrecan is cleaved within the interglobular domain of aggrecan at site Glu(373)-Ala(374). Aggrecan degradation has been linked to the development of arthritic diseases such as rheumatoid arthritis and osteoarthritis as well as retinal disease. Alternate Names: ADAMTS5, ADAMTS11, ADMP2, ADAMTS-5, ADAM-TS 5, ADAM-TS5, Aggrecanase-2, ADMP-2, ADAMTS-11, ADAM-TS 11, A disintegrin and metalloproteinase with thrombospondin motifs 5, A disintegrin and metalloproteinase with thrombospondin motifs 11.
Specificity This antibody is specific for ADAMTS5 and shows no cross-reactivity with other ADAMTS family members.

Application Details

Application Notes Western blotting (1 ug/ml): Detects a major band at ~68 kDa protein representing ADAMTS5 from porcine cartilage extract. On human recombinant ADAMTS5 this antibody detects a major band at ~85 kDa. Depending on the extent of C-terminal processing, several smaller bands may also be seen using this antibody. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user. Further Comments: Expected to crossreact with mouse due to sequence identity. This polyclonal antibody has been referred to as: JSCKNG.
Concentration 1.0 mg/ml
Buffer PBS containing 1 mg/ml BSA as stabilizer and 0.05% Sodium Azide as preservative
Storage Store the antibody undiluted (in aliquots) at -20°C. Avoid repeated freezing and thawing. Shelf life: one year from despatch.
Research Area Proteolysis / Ubiquitin, Metalloprotease, Extracellular Matrix, Proteases
Restrictions For Research Use only
