Additional Information

Alternative name MLH-1
Immunogen Full length recombinant MLH.
Isotype IgG1  (Matching secondary antibodies)
Clone G168-15
Specificity The G168-15 antibody recognizes the human and mouse MLH1 (80-85kDa. The repair of mismatch DNA is essential to maintaining the integrity of genetic information over time. An alteration of microsatellite repeats is the result of slippage owing to strand misalignment during DNA replication and is referred to as microsatellite instability (MSI). These defects in DNA repair pathways have been related to human carcinogenesis. The importance of mismatch repair genes became apparent with the identification of the genetic basis for hereditary nonpolyposis colon cancer (HNPC). MSH-2 is involved in the initial cognition of mismatch nucleotides during the replication mismatch repair process. It is thought that after MSH2 binds to a mismatched DNA duplex it is joined by a heterodimer of MLH1 and PMSH, which together help facilitate the later steps in mismatch repair. This antibody cross reacts with human, hamster and rat

Application Details

Application Notes Titer/Working Dilution: This antibody may be diluted to a titer of 1:25-1:50 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. Staining Protocol: We suggest an incubation period of 30 to 60 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Positive Control: Tonsil, colon carcinoma. Cellular localization: Nuclear
Concentration 100ug/ml approximately
Buffer This antibody is supplied as liquid containing sodium azide as preservative.
Storage Store the antibody at 2-8o c. Do not use it beyond the expiration date stated on the label.
Restrictions For Research Use. CE-certified for In Vitro Diagnostics (IVD) Use in the European Union
