Specificity

This antibody stains with a 300 kD protein. P300 and CBP (CREB-binding protein) are homologous to each other. P300 is transcriptional adapter protein, which is characterized by three cysteine and histidine-rich regions. Protein kinase A mediated CREB phosphorylation results in the binding of CREB to a 265 kD nuclear protein designated CBP. This antibody crossreacts with monkey, mink, mouse and rat.

Application Notes

Titer/Working Dilution: This antibody may be diluted to a titer of 1:25-1:50 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. Staining Protocol: We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Positive Control: Tonsil.
