Specificity P504S has been recently described as prostate cancer-specific gene that encodes a protein involved in the beta-oxidation of branched chain fatty acids. Expression of P504S protein is found in prostatic adenocarcinoma but not in benign prostatic tissue. It stains premalignant lesions of prostate: high-grade prostatic intraepithelial neoplasia (PIN) and atypical adenomatous hyperplasia. P504S can be used as a positive marker for PIN. The p63 gene is highly expressed in the basal or progenitor layers of many epithelial tissues. P63 shows remarkable structural similarity to p53 gene. Unlike p53, the p63 gene encodes multiple isotypes with remarkable divergent abilities to transactivate p53 reporter genes and induce apoptosis. The combination of P504S and p63 (PIN) may be extremely useful for diagnosing prostatic intraepithelial neoplasia (PIN). P504S stains cytoplasm in prostatic adenocarcinomas and atypical hyperplasia and p63 stains nuclei in normal and benign prostate glands.

Application Details

Application Notes Titer/Working Dilution: This antibody may be diluted to a titer of 1:25-1:50 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. Staining Protocol: We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Positive Control: Prostate carcinoma.
