Specificity
|
This antibody reacts specifically with human ET-1. The antibody shows 90% crossreactivity with vasointestinal constrictor, <10% crossreactivity with ET-2 and ET-3, and no cross-reactivity with sarafotoxin 6c, the C-terminal hexapeptide of ET-1. The Endothelins show potent constrictor activity in vascular and non-vascular smooth muscle. This family of 21-amino acid peptides exists in at least three isoforms - ET-1, ET-2, and ET-3, and is produced in endothelial and epithelial cells.

Application Notes

Titer/Working Dilution: This antibody may be diluted to a titer of 1:50 to 1:100 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. Staining Protocol: We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Positive Control: Placenta, Lung.
