This antibody reacts with a 65 kD protein. NF kappa B is a heterodimer of a 50 kD DNA binding subunit and a 65kD transactivating subunit (RelA). The p50/p65 heterodimer is present in the cytoplasm in inactive form bound to its inhibitor, IkB. In unstimulated cells, nuclear localization signals present on p65 are masked by members of the IkB family of inhibitory proteins. Subsequent to cell stimulation, IkB undergoes phosphorylation, allowing nuclear translocation of the active dimeric NFkB transcription factor. This antibody crossreacts with rat and rabbit.

Application Notes

Titer/Working Dilution: This antibody may be diluted to a titer of 1:25-1:50 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. Staining Protocol: We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation conditions should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Positive Control: Prostate, placenta.
