Description
Cytokeratin 1 is a type II Cytokeratin. The type II Cytokeratins consist of basic or neutralproteins which are arranged in pairs of heterotypic keratin chains coexpressed duringdifferentiation of simple and stratified epithelial tissues. This type II Cytokeratin isspecifically expressed in the spinous and granular layers of the epidermis with familymember KRT10 and mutations in these genes have been associated with bullouscongenital ichthyosiform erythroderma.
Specificity
Reacts with Basic keratin K1 (Mr 66 000). Specific for keratin K1 in suprabasal layers of cornified squamous epithelia, e. g. thesuprabasal layer of scalp and foot sole. Negative with non-keratinizing stratified and non-stratified epithelia (e. g. esophagus, duodenum)! Species Reactivity: Tested: Human and Mouse.
Application Notes
Cytological Material. Western blotting: 1/3,000 (ECL). Immunohistochemistry on Frozen Sections: 1/200. Immunohistochemistry on Paraffin Embedded Material: 1/100 (after microwave treatmentor trypsin). Incubation Time: 1 h at RT, extended with paraffin sections. Positive Control: Human foot sole. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
