Description
|
Cytokeratin 20 is a type I keratin which is primarily expressed in gastric and intestinalepithelium, urothelium, and Merkel-cells. The keratins are intermediate filament proteinsresponsible for the structural integrity of epithelial cells and are subdivided intocytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which arearranged in pairs of heterotypic keratin chains. Their genes are clustered in a region ofchromosome 17q12-q21. Cytokeratin 20 is a major cellular protein of mature enterocytesand goblet cells and is specifically expressed in the gastric and intestinal mucosa. It isalso expressed in adenocarcinomas of the colon, stomach, pancreas and the bile systemand is present in mucinous ovarian tumors, transitional-cell and Merkel-cell carcinomas. Notably, the squamous cell carcinomas and adenocarcinomas of the breast, lung, andendometrium, non-mucinous tumors of the ovary, and small cell carcinomas lackcytokeratin 20.

Specificity
|
Reacts with acidic human keratin K20 (Mr 46 000). Excellent marker for certain types of carcinomas such as adenocarcinomas of the colon,transitional cell carcinomas of the bladder and Merkel cell tumors of the skin. Very sensitive detection of intestinal and gastric foveolar epithelium, urothelial umbrellacells, Merkel cells of epidermis as well as tumors originating therefrom (e. g. primary andmetastatic colorectal carcinoma).

Western Blot: 1/3000 (ECL). Immunohistochemistry on Frozen Sections: 1/200. Immunohistochemistry on Paraffin Embedded Tissue: 1/100 (after microwave treatment ortrypsin). Cytological material. Incubation Time: 1 h at RT, extended with paraffin sections. Positive Control: Colon, duodenum, Merkel cells of skin. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
