Description BRCA1-8F7 recognizes full-length BRCA1, a 220kDa nuclear phosphoprotein. In a high proportion of breast and ovarian cancer cell lines, BRCA1 aberrantly mislocates to the cytoplasm. Its usefulness to monitor functional inactivation of BRCA1 in sporadic breast cancers is under active investigation.

Application Notes For WB: Use at a concentration of 1-10ug/ml. This antibody recognizes full-length BRCA1, a 220-kDa nuclear phosphoprotein. For IP: Use at an assay dependent dilution. For IHC: Use at an assay dependent dilution. Block endogenous peroxidase with 0.3% H2O2 for 30 minutes. Perform heat mediated antigen retrieval before commencing with IHC staining protocol (0.1 M citrate buffer at 95C in water bath for 20 min). Avoid nonspecific binding by incubating in normal rabbit anti-mouse serum. Incubate primary antibody overnight and use a biotin-streptavidin amplification kit. Not tested in other applications. Optimal dilutions/concentrations should be determined by the researcher.
