Background Rad50, a 153kDa protein, is associated with Mre11 and Nbs1 (Nibrin or p95) to form amultiprotein complex involved in the double strand break repair process. Rad50 is amember of the family of structural maintenance of chromosome (SMC) proteins. Members of this family share similar structures consisting of globular N- and C-terminiseparated by a coiled-coil domain. At the base of the coiled-coil region is a zinc bindingmotif, the zinc hook. The zinc hook mediates Rad50 dimerization through the linkage oftwo Rad50 coiled-coil tails. Protocol Western blot protocol specific for RAD50 Antibody Western blot
1. Gel/Transfer set up according to manufacturer's instructions (NuPAGE, Invitrogen).
. Block: 1X TBST +5 % milk (TBST: 100mM Tris pH 7.5, 150mM NaCl, 0.1 % Tween) for 1 hour at room temperature.
. Wash 3 times (5 minutes) in 1X TBST.
. Primary Antibody: Rad50, hook serum diluted 1:5,000 in 1X TBST and incubated for 1 hour at room temperature.
. Wash 3 times (10 minutes) in 1X TBST.
. Secondary Antibody: Anti-rabbit IgG diluted in 1X TBST and incubated for 1 hour at room temperature.
. Wash 3 times (5 minutes) in 1X TBST.
. Expose: Chemiluminescent HRP for 15-60 seconds. NOTE: This is the protocol used to obtain the image on the datasheet.
