Immunogen N-terminal fragment of human 14-3-3 gamma
Clone HS23
Isotype IgG1 kappa
Specificity NB100-407 reacts with the human form of intact, non-modified N-terminus 14-3-3 gamma. The naturally occurring form of 14-3-3 gamma in which the N-terminus Met is removed and the Val is acetylated is not recognized by this antibody.
Purification Ascites
Alternative Name 14-3-3 protein gamma
Background 14-3-3 proteins are present as multigene families in most organisms. Signal-inducedphosphorylation has the ability to change protein function. Sometimes, however, phosphorylation is not enough to change a protein',s function. 14-3-3 proteinsplay an important role of bringing signal transduction to completion. They regulate manycellular processes that are important in cancer biology, such as apoptosis and cell-cyclecheckpoints. Alternate Names: anti-KCIP-1 antibody, anti-KCIP1 antibody, anti-KCIP 1 antibody, anti-14 3 3 proteingamma antibody, anti-14 3 3gamma antibody, anti-14-3-3 gamma antibody, anti-14 33gamma antibody, anti-14-3-3 protein gamma subtype antibody, anti-3monooxygenase/tryptophan 5 monooxygenase activation protein gamma polypeptideantibody, anti-Protein kinase C inhibitor protein 1 antibody, anti-Tyrosine 3monooxygenase/tryptophan 5 monooxygenase activation protein gamma polypeptideantibody, anti-YWHAG antibody.
Gene Symbol: YWHAG
Gene ID 7532
UniProt P61981
Application Notes This 14-3-3 gamma (HS23) antibody can be used in Immunocytochemistry/Immunofluorescence and Western Blot, where a distinct band around 33 kDa is detected.
Recommended dilutions: Immunocytochemistry/Immunofluorescence 1:50-1:250, Western Blot 1:500-1:1000
Protocol Western Blot protocol specific for 14-3-3 gamma Antibody
1. Load protein on gel (ie: ~30 µg of HeLa whole cell control lysate) and run.
. Transfer protein to nitrocellulose (Schleicher&Schuell, cat# BA83).
. Block the membrane with 5 % milk for 30 minutes at room temperature.
. Incubate the membrane with anti-14-3-3 gamma [cat# NB 100-407] (1:1,000), overnight at 4C or 2 h at room temperature. Milk or PBS-T are good for this dilution.
. Wash the membrane 3 times, 10 minutes per wash in PBS with 0.1 % Tween-20 (PBS-T).
. Incubate with secondary antibody for 30 minutes at room temperature. Use milk in PBS-T as a diluent.
. Rinse the membrane 2 times with deionized water and place in an ECL working solution.
. Expose to the film.
Restrictions For Research Use only
Format Liquid
Preservative Sodium azide
Precaution of Use WARNING: Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
Handling Advice Avoid freeze-thaw cycles
Storage 4 °C
Storage Comment 4 °C short term. Aliquot and store at -20 °C long term.
