Immunogen A synthetic peptide within the extreme C-terminus of human FANCJ.
Purification Whole antisera
Alternative Name BRIP1 / FANCJ
Background BRIP1 (also called BACH1 /FANCJ) is a helicase that interacts with the BRCT domainof BRCA1 and has a role in BRCA1-dependent DNA repair and checkpoint. BRIP1 hasrecently been found to be defective in Fanconi anemia complementation group J. BRIP1/FANCJ has a function in the Fanconi anemia pathway that is independent ofBRCA1 and downstream of FANCD2 activation. (1-4). Alternate Names: anti-FANCJ protein antibody, anti-BACH 1 antibody, anti-BACH1 antibody, anti-BRIP1antibody, anti-BRIP 1 antibody, anti-BRAC 1 Associated C Terminal Helicase 1antibody, anti-BRCA 1 Interacting Protein 1 antibody, anti- Fanconi anemia group Jprotein.
Gene Symbol: BRIP1
Gene ID 83990
UniProt Q9BX63
Application Notes This FANCJ antibody is useful for Immunohistochemistry on paraffin-embedded sections, Immunoprecipitation and Western blot, where a band can be seen at ~130 kDa. NB100-416 has also been successfully used for Immunofluorescence as reported by a customer review.
Recommended dilutions: Immunocytochemistry/Immunofluorescence, Immunohistochemistry 1:10-1:2000, Immunohistochemistry-Paraffin 1:200, Immunoprecipitation 1:10-1:500, Western Blot 1:500-1:200
Protocol Chromatin Immunoprecipitation (ChIP) protocol specific for FANCJ Antibody 1) Grow cells in Dulbecco's Modified Eagle Medium (DMEM) supplemented with 10 % fetal bovine serum to 95 % confluence. 2) Cross-linked cells with 1 % formaldehyde at room temperature for 10 minutes. 3) Rinse with ice-cold PBS twice and collect into 100 mM Tris-HCl (pH 9.
4) and 10 mM DTT.
) Incubate for 15 minutes at 30C and centrifuge for 5 minutes at 2,000 rpm.
) Wash cells sequentially with: a. 1.0 mL ice-cold PBSb.
Restrictions For Research Use only
Format Liquid
Buffer I [0.25 % Triton X-100, 10 mM EDTA, 0.5 mM EGTA, 10 mM HEPES, pH 6.5)c, Sodium Azide
Preservative Sodium azide
Precaution of Use WARNING: Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
Handling Advice Avoid freeze-thaw cycles
Storage 4 °C
Storage Comment 4 °C short term. Aliquot and store at -20 °C long term.
