Background BRIP1 (also called BACH1) is a helicase that interacts with the BRCT domain of BRCA1and has a role in BRCA1-dependent DNA repair and checkpoint. BRIP1/FANCJhas a function in the Fanconi anemia pathway that is independent of BRCA1 anddownstream of FANCD2 activation. (1-4). Application Notes This FANCJ antibody is useful for Immunohistochemistry on paraffin-embedded sections, Immunoprecipitation and Western blot, where a band can be seen at ~130 kDa. NB100-416 has also been successfully used for Immunofluorescence as reported by a customer review.
Recommended dilutions: Immunocytochemistry/Immunofluorescence, Immunohistochemistry 1:10-1:2000, Immunohistochemistry-Paraffin 1:200, Immunoprecipitation 1:10-1:500, Western Blot 1:500-1:200
Protocol Chromatin Immunoprecipitation (ChIP) protocol specific for FANCJ Antibody 1) Grow cells in Dulbecco's Modified Eagle Medium (DMEM) supplemented with 10 % fetal bovine serum to 95 % confluence. 2) Cross-linked cells with 1 % formaldehyde at room temperature for 10 minutes. 3) Rinse with ice-cold PBS twice and collect into 100 mM Tris-HCl (pH 9.
4) and 10 mM DTT.
) Incubate for 15 minutes at 30C and centrifuge for 5 minutes at 2,000 rpm.
) Wash cells sequentially with: a. 1.0 mL ice-cold PBSb.
