Immunogen A synthetic peptide made to the C-terminal region of human Derlin 1 (between residues 200-251).
Cross-Reactivity (Details) Predicted to react with Xenopus, bovine, chicken, rat, zebrafish, mouse based on 100% sequence homology.
Purification affinity purified
Alternative Name Derlin-1
Background Derlin-1 is a channel-like protein, found in the ER membrane, responsible for degradingand funneling misfolded proteins and other cellular debris into the appropriatedegradative pathways within the cell. Alternate Names: anti-Degradation in endoplasmic reticulum protein 1 antibody, anti-Der1-like protein 1antibody, anti-DERtrin-1 antibody, anti-DERL1 antibody.
Gene Symbol: DERL1
Gene ID 79139
UniProt Q9BUN8
Application Notes This Derlin 1 antibody can be used for Immunocytochemistry/Immunofluorescence, Immunohistochemistry on paraffin-embedded sections and Western blot, where a band is seen at ~29 kDa. In ICC/IF, ER staining was observed in HeLa cells.
Recommended dilutions: Immunocytochemistry/Immunofluorescence 1:2000, Immunohistochemistry 5-10 µg/mL, Immunohistochemistry-Paraffin 5-10 µg/mL, Western Blot 1:1000-1:5000
Protocol Protocol specific for Derlin 1 Antibody Western Blot
1. Run samples on a 10-15 % SDS-PAGE.
. Transfer proteins to a methanol soaked PVDF membrane for 2.5 hr at 250 milliamper at 4C on a stir plate.
. Immerse PVDF membrane in blocking buffer and block for 1 hour at room temperature.
. Wash membrane 3 times with TBS-T.
. Incubate with primary Derlin-1 antibody , diluted in blocking buffer 1:500, overnight at 4C
. Wash membrane 3 x 10 min with TBS-T.
. Incubate with secondary antibody, diluted in blocking buffer, at room temp for 1 hour.
. Wash 3 x 10 min with TBS-T.
. Detect with Amersham ECL kit (RPN 2106).
Restrictions For Research Use only
Format Liquid
Concentration 1 mg/mL
Buffer Tris-citrate/Phosphate, pH 7-8, Sodium Azide
Preservative Sodium azide
Precaution of Use WARNING: Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
Handling Advice Do not freeze.
Storage 4 °C
