Background P300 is a relatively stable, ubiquitously expressed, nuclear phosphoprotein which isconserved among a variety of mammalian species. It is actively synthesized andphosphorylated in both quiescent and proliferating cells.Cyclic AMP-responsive enhancer binding protein (CREB) binding protein (CBP) andp300 are closely related transcriptional coactivators and have been shown to directlyinteract with many different DNA-binding transcription factors including nuclear hormonereceptors, CREB, c-Fos, C-Jun/v-Jun, c-Myb/v-Myb, TFIIB and MyoD. Both CBP andp300 have been shown to display histone acetyltransferase (HAT) activity, capable ofacetylating all four core histone particles in nucleosomes. As a result of HAT activity, ithas been suggested CBP and p300 may play a direct role in activating chromatin fortranscription. Alternate Names: anti-CREBBP/EP300 inhibitory protein 1 antibody, anti-Cyclic AMP responsive enhancerbinding protein antibody, anti-EP300: E1A binding protein p300 antibody, anti-RB andP300 binding protein EID 1 antibody, anti-Retinoblastoma protein associated proteinantibody.

Application Notes This KAT3B/p300 antibody is useful for Immunocytochemistry/Immunofluorescence, Immunoprecipitation and Western Blot. In Western blot a band is seen at ~300 kDa, representing the p300 protein. Lower non-specific MW bands (~45-66 kDa) ay be seen with longer exposure times.
Recommended dilutions: Chromatin Immunoprecipitation, Immunocytochemistry/Immunofluorescence 1:50-1:200, Immunoprecipitation 1:10-1:500, Western Blot 1:250-1:500
Protocol Protocol specific for KAT3B / p300 Antibody Western Blot Protocol
1. Perform SDS-PAGE (3-8 % Tris-acetate) on samples to be analyzed, loading 50ug of total protein per lane.
. Transfer proteins to Nitrocellulose according to the instructions provided by the manufacturer of the transfer apparatus.
. Stain the blot using ponceau S for 1-2 minutes to access the transfer of proteins onto the nitrocellulose membrane. Rinse the blot in water to remove excess stain and mark the lane locations and locations of molecular weight markers using a pencil.
. Rinse the blot in TBS for approximately 5 minutes.
. Block the membrane using 5 % non-fat dry milk in TBS for 1.5 hours.
. Dilute the mouse anti-p300 primary antibody in blocking buffer and incubate overnight at 4 °C.
. Wash the membrane in water for 5 minutes and apply the diluted mouse-IgG HRP-conjugated secondary antibody in blocking buffer (as per manufacturer's instructions) and incubate 1 hour at room temperature.
. Wash the blot in TBS containing 0.05-0.1 % Tween-20 for 10-20 minutes.
. Wash the blot in type I water for an additional 10-20 minutes (this step can be repeated as required to reduce background).
. Apply the detection reagent of choice in accordance with the manufacturer's instructions (Amersham's ECL is the standard reagent used).Note: Tween-20 can be added to the blocking buffer at a final concentration of 0.05-0.2 %, provided it does not interfere with antibody-antigen binding.
