Immunogen ABIN152549 was raised against a synthetic phosphopeptide corresponding to aminoacid residues surrounding the phosphorylated Thr305 of CAM KII. This sequence isidentical in human, rat, mouse, drosophila, and limulus and in alpha, beta, and deltaforms.
Specificity ABIN152549 is specific for CAM KII phosphorylated at Thr305. The labeling of the CAMKII bands in Western Blots of rat brain is blocked by the phosphopeptide but not by thedephosphopeptide.Phosphorylated:Thr305. Species Reactivity: Detects rat, mouse, human, bovine, chicken and xenopus. Immunolabeling is absent inhomogenate of brain from T305D mutant mice.

Background CaM KII is an important member of calcium/calmodulin-activated kinase, functioning inneural synaptic stimulation and T-cell receptor signaling. CaM KII has catalytic andregulatory domains. The binding of Ca++/calmodulin to its regulatory domain releasesits autoinhibitory effect and activates the kinase. The activated CaM KII furtherautophosphorylates itself at Thr286 to render the kinase constitutively active. Thethreonine phosphorylation state of CaM KII can be regulated through PP1/PKA. PP1(protein phosphatase 1) dephosphorylates phospho-CaM KII at Thr286. On the otherhand, PKA (protein kinase A) prevents this phosphorylation process through itsinhibitory effect on PP1.
