Specificity Specific for the ~83k B-catenin phosphorylated at Ser33,37 in Western blots of humanembryonic kidney cells. Phosphorylated:Ser33/Ser37. Species Reactivity: Recognizes human and rat forms of B-catenin. Not tested in other species. Thesequence of the immunogen is identical in human, mouse, rat and Xenopus. Localization: Cytoplasmic (high level of ser/thr phosphorylation) or bound to CDH1. Translocates tothe nucleus (low level of ser/thr phosphorylation).

Background B-Catenin is a central component of the cadherin cell adhesion complex and plays anessential role in neural development in the Wingless/Wnt signaling pathway (Ding andDale, 2002, Polakis, 2000). The role of B-catenin in these processes is thoughtregulated by the sequential phosphorylation of Ser29 Ser33, Ser37 and Thr41 byglycogen synthase kinase 3B (GSK3B) (Liu et al., 2002). This hyperphosphorylationpromotes the ubiquitylation and targeted destruction of B-catenin. Mutations incomponents of this phosphorylation regulated process that prevent B-cateninhyperphosphorylation by GSK3B are strongly associated with cancers (Wang et al.,2003, Polakis, 2000, Liu et al., 2002). Alternate Names: anti-Beta-Catenin antibody, anti-B-Catenin antibody, anti-CTNNB antibody.
