Background Steroid and thyroid hormones and retinoic acid regulate a complex array of geneexpression activity via intracellular receptor transcription factors belonging to theligand-dependent nuclear receptor superfamily. Adding to the complexity of function ofthese transcription factors are associated proteins known as coactivators andcorepressors which, as their names suggest, enhance or depress transcription activity ofthe nuclear receptor with which they associate. Amplified in breast cancer-1 (AIB-1), is amember of a growing family of transcriptional coactivator proteins which includes steroidreceptor coactivator-1 (SRC-1) and glucocorticoid receptor interacting protein-1(GRIP1).AIB1/RAC3 has been shown to interact and stimulate transcriptional activity of theretinoic acid, retinoid X, vitamin D, mineralocorticoid, glucocorticoid, thyroid hormone,androgen, progesterone, and peroxisome proliferator activated receptors. AIB1/RAC3also interacts with the transcriptional integrators CREB binding protein (CBP) and p300.
