Clone 3H2-E8
Isotype IgG1
Purification Protein G purified
Alternative Name BLIMP1/PRDM1 (PRDM1 Antibody Abstract)
Background Gene Symbol: PRDM1
Gene ID 639
UniProt Q60636
Pathways Regulation of Muscle Cell Differentiation
Application Notes Western Blot 1:1000, Chromatin Immunoprecipitation 1 μg antibody/ 10 μg protein, Flow Cytometry 8 μg/mL where cells are used at 2*10^6/mL, ELISA 1:100-1:2000, Immunohistochemistry 1:200, Immunocytochemistry/Immunofluorescence 1:50-1:200, Immunoprecipitation, Immunohistochemistry-Paraffin 1:200, Immunohistochemistry-FrozenThis Blimp-1 (3H2-E8) antibody is useful for ELISA, Immunocytochemistry/Immunofluorescence, Western Blot, Immunohistochemistry on paraffin-embedded sections, Flow Cytometry, Immunoprecipitation and Chromatin Immunoprecipitation applications. Immunohistochemistry-Frozen was reported in scientific literature. By WB, this antibody recognizes a band at ~98 kDa and may recognize one at ~80 kDa (the beta form). Antigen retrieval is recommended (EDTA buffer, microwave) prior to IHC on paraffin tissues. This antibody demonstrates nuclear staining. For IHC and ICC/IF a dilution of 1:200 is recommended with tyramide amplification.
Comment

The antibodies are intended for use in vitro experiments only. Our antibodies have not been tested nor are recommended for use in vivo.

Protocol Protocol specific for Blimp-1 Antibody Western Blot
1. Run ~20 micrograms of protein on a 10 % SDS-PAGE.
. Block nitrocellulose membrane with 5 % blotto in TBS-0.2 % Tween-20, overnight at 4 degrees Celcius.
. Incubate membrane with diluted anti-Blimp-1 (primary antibody, cat# NB 600-235) for 1 hour at RT, in 2 % blotto in TBS-0.2 % Tween-20.
. Wash blot 4 times for 10 minutes, each, at RT with TBS-0.2 % Tween-20.
. Incubate membrane with anti-mouse IgG1 conjugated to HRP (secondary antibody), diluted in 2 % blotto in TBS-0.2 % Tween-20, for 45 minutes at RT.
. Wash 4 times for 10 minutes, each, at RT with TBS-0.2 % Tween-20.
. Incubate membrane with chemiluminescence reagent as per vendor specification. Dry blot and cover with saran wrap.
. Expose to film.9.Develop image.Immunohistochemistry / ImmunofluorescencePlease refer to the following refernence:Cattoretti, G, et al. PRDM1/Blimp-1 is expressed in human B-lymphocytes committed to the plasma cell lineage.J Pathol. 206(1):76-86 (2005)
