Immunogen A fusion protein containing amino acids 284-709 of human Nucleolin.
Purification Whole antisera
Alternative Name Nucleolin (NCL Antibody Abstract)
Background Nucleolin is an abundant, 106 kDa nucleolar phosphoprotein. It is the major protein inactively dividing cells, whereas the degraded forms are relatively abundant innon-dividing cells. Stability of the nucleolin protein is cell proliferation-dependent. Itplays a role in rDNA transcription, organization, and rRNA processing. It may alter DNAorganization in response to cell cycle controls. It is also a good mammalian nucleolusmarker. Alternate Names: anti-nucl antibody, anti-protein C23 antibody, anti-NCL antibody, anti-Nucleolin antibody,anti-C23 antibody.
Gene Symbol: NCL
Gene ID 4691, 17975
UniProt P19338
Research Area Chromatin and Nuclear Signaling, DNA/RNA, Transcription Factors
Application Notes This Nucleolin antibody is useful for Western blot, Immunocytochemistry/Immunofluorescence, Immunohistochemistry on paraffin-embedded sections and Electron Microscopy. Chromatin Immunoprecipitation was reported in scientific literature. By Western Blot, a band at ~106 kDa represents human Nucleolin. Heat mediated antigen retrieval is recommended for IHC-P.
Recommended dilutions: Chromatin Immunoprecipitation, Electron Microscopy 1:10-1:500, Immunocytochemistry/Immunofluorescence 1:50-1:100, Immunohistochemistry 1:50-1:100, Immunohistochemistry-Paraffin 1:50-1:100, Immunomicroscopy, Western Blot 1:4000
Protocol Protocol specific for Nucleolin Antibody IHC-FFPE sections I. Deparaffinization: A. Treat slides with Xylene: 3 changes for 5 minutes each. Drain slides for 10 seconds between changes. B. Treat slides with 100 % Reagent Alcohol: 3 changes for 5 minutes each. Drain slides for 10 seconds between changes. II. Quench Endogenous Peroxidase: A. Place slides in peroxidase quenching solution: 15-30 minutes. To Prepare 200 mL of Quenching Solution: Add 3 mL of 30 % Hydrogen Peroxide to 200 mL of Methanol. Use within 4 hours of preparation B. Place slides in distilled water: 2 changes for 2 minutes each. III. Retrieve Epitopes: A. Preheat Citrate
Restrictions For Research Use only
Format Liquid
Preservative Sodium azide
Precaution of Use WARNING: Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
Handling Advice Avoid freeze-thaw cycles
Storage -20 °C
Storage Comment Aliquot and store at -20 °C or -80 °C.
