Immunogen slightly modified beta-cytoplasmic actin N-terminal peptide, Ac-Asp-Asp-Asp-Ile-Ala-Ala-Leu-Val-Ile-Asp-Asn-Gly-Ser-Gly-Lys, conjugated to KLH.
Clone AC-15
Isotype IgG1
Specificity In staining of chicken gizzard ultrathin tissue cryosections, the antibody labels the dense bodies and longitudinal channels linking consecutive dense bodies that are also occupied by desmin and the membrane-associated dense plaque. It does not stain adult cardiac and skeletal muscles except for traces due to contaminations of the sample with non-muscle cells, or if embryonic tissue is being used. The antibody can be used for staining of acetone-fixed frozen sections, EM preparations, and microinjection experiments. B5, ethanol, methacam, or Bouin's solutions can be used as fixatives. The epitope recognized by the antibody is resistant to formalin-fixed and paraffin-embedding.
No Cross-Reactivity Fruit Fly (Drosophila melanogaster)
Cross-Reactivity (Details) Does not cross react with adult cardiac, skeletal muscle, drosophila, amoeba beta actin. Hamster reactivity reported in scientific literature (PMID: 24478435) Not yet tested in other species.
Purification Ascites
Alternative Name Actin beta / ACTB
Background Beta-actin is one of six different actin isoforms which have been identified. Actins arehighly conserved proteins that are involved in cell motility, structure, and integrity.Because beta-actin is ubiquitously expressed in all eukaryotic cells, it is frequently usedas a loading control for assays involving protein detection, such as Western blotting.Antibodies to beta-actin provide a specific and useful tool in studying the intracellulardistribution of beta-actin and the static and dynamic aspects of the cytoskeleton. Alternate Names: Anti-A X actin like protein antibody, Anti-Actx antibody, Anti-Beta cytoskeletal actinantibody, Anti-Melanoma X actin antibody, Anti-PS1TP5BP1 antibody, Anti-Beta-actinantibody, Anti-Beta actin antibody, Anti-Actin, beta antibody.
Gene Symbol: ACTB
Gene ID 60, 11461, 81822
Application Notes This antibody is useful for Western blot, ELISA, immunohistochemistry and Immunocytochemistry. Molecular weight of beta actin is 42kD. The antibody can be used for staining of acetone-fixed frozen sections, EM preparations, and microinjection experiments. This antibody is not suitable for formalin fixation and paraffin embedding. B5, methacarn, ethanol or Bouin's solutions may also be used as fixatives.
Recommended dilutions: ELISA 1:100-1:2000, Immunocytochemistry/Immunofluorescence 1:1000-1:2000, Immunohistochemistry 1:10-1:500, Immunohistochemistry-Frozen 1:500, Immunohistochemistry-Paraffin (Negative), Western Blot 1:5000-1:10000
Protocol Protocol specific for Beta Actin Antibody IHC-FFPE sections:I. Deparaffinization:A. Treat slides with Xylene: 3 changes for 5 minutes each. Drain slides for 10 seconds between changes. B. Treat slides with 100 % Reagent Alcohol: 3 changes for 5 minutes each. Drain slides for 10 seconds between changes.II. Quench Endogenous Peroxidase:A. Place slides in peroxidase quenching solution: 15-30 minutes. To Prepare 200 mL of Quenching Solution: Add 3 mL of 30 % Hydrogen Peroxide to 200 mL of Methanol. Use within 4 hours of preparation B. Place slides in distilled water: 2 changes for 2 minutes each. III. Retrieve Epitopes:A. Preheat Citrate
Restrictions For Research Use only
Format Liquid
Buffer Ascitic fluid, Sodium Azide
Preservative Sodium azide
Precaution of Use WARNING: Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
Handling Advice Avoid freeze-thaw cycles
Storage 4 °C
Storage Comment 4 °C short term. Aliquot and store at -20 °C long term.
