Background Human malignant neoplasms carry rejection antigens that are recognized by the patients'autologous, tumor directed and specific, cytolytic, CD8+ T lymphocyte clones (CTL). Animportant group of antigens is coded by the MAGE family of genes. It was identified thatmelanomas and primary glial brain tumors express common melanoma associatedantigens (MAAs). Because MAGE-1 is expressed on a significant proportion of humanneoplasms of various histological types (melanoma, brain tumors of glial origin,neuroblastoma, nonsmall cell lung cancer, breast, gastric, colorectal, ovarian, renal cellcarcinomas) and not on normal tissues, the encoded antigen may serve as a marker ofearly detection and target for cancer immunotherapy.

Application Notes Immunofluorescence. Western Blotting: 0.5-1.0 ug/ml for 2 hrs at RT. Immunohistology (Formalin/paraffin only): use 4-8 ug/ml for 30 min at RT, staining offormalin-fixed tissues REQUIRES boiling tissue sections in 1 mM EDTA, pH 8.0, for 10-20minfollowed by cooling at RT for 20 min. Positive Control: Colo32 cells Germ cells in testis. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
