Immunogen A synthetic 19-mer peptide, corresponding to aa 8-26 (VKPPQNKTESENTSDKPKR) from thesecreted amphiregulin of human origin.
Description Amphiregulin (AR) binds to EGF-Receptor (EGFR) with lower affinity than EGF. The maturesecreted form AR is an 84-amino acid residue glycosylated polypeptide growth regulator,which is generated by proteolytic processing of a 252-amino acid transmembraneprecursor. Seven different polypeptide ligands, which derive from distinct genes, arecapable of binding to the extracellular domain of EGFR. These ligands include EGF,TGFalpha, AR, HB-EGF, cripto-1, epiregulin, and betacellulin. All of these growth factorscontain a characteristic EGF-like domain which is defined by 6 evenly spaced cysteineresidues that generate 3 loops through the formation of disulfide bonds. AR protein hasbeen localized by immunohistochemistry to the epithelium of the colon, stomach,pancreas, breast, and placenta. AR is reportedly overexpressed in human cancers ofbreast, colon, stomach, and pancreas.
Specificity Highly specific for Amphiregulin. Cellular Localization: Cytoplasmic and/or nuclear. Mol. Wt. of Antigen: 18. 5-22. 5kDa. Species: Human and Mouse. Others not tested.

Application Details

Application Notes Immunoprecipitation (Native): use Protein A and Ab at 10 g/mg protein lysate. PositiveControl: LS174T cells. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
Buffer 10mM PBS, pH 7.4, with 0.2% BSA and 0.09% sodium azide
