Background Caspase 8 binds to the death effector domain (DED) of FADD through an analogous DED domain present in tandem in the proform of the Caspase 8 protein. Recruitment of Caspase 8 to the Fas receptor results in oligomerization of the Caspase 8 protein, which in turn drives its autoactivation through “self-cleavage”. Activated Caspase 8 then activates other downstream caspases including Caspase 9, thereby commiting the cell to undergo apoptosis.

Application Notes Immunoprecipitation (10 g/mg protein lysate): Denatured verified, use Protein A. Western Blotting (2.5-5 ug/ml for 2hrs at RT). Immunohistology (Formalin/paraffin9: 5-10 ug/ml for 30 min at RT. Staining offormalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for10-20 min followed by cooling at RT for 20 min. Recommended positive control: Jurkat cells, Stomach. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
