Description
Bub1 cooperates with Aurora B kinase to inhibit Cdc20-mediated activation of theanaphase promoting complex/cyclosome (APC/C) until all the chromosomes correctly alignon the microtubule spindle apparatus via their kinetochores, thus ensuring high fidelitymitotic chromosome segregation. Bub1 is also required for assembly of a functional innercentromere, sister chromatid cohesion via targeting of the Shugoshin protein, andmetaphase congression. Mutation or aberrant Bub1 expression is associated withchromosomal instability, aneuploidy, and human cancer.
Specificity
This antibody detects Bub1, a protein kinase involved in spindle checkpoint functionduring mitosis. Species: Human. Others not tested.

Application Notes
Immunoprecipitation: 1: 200. Western Blot: 1: 500 - 1: 2000, detects a band of approximately 160 kDa in HeLa cell lysates. Immunofluorescence: 1: 500 - 1: 2000 (use cells grown on cover slips fixed with 3.5%paraformaldehyde in PBS at pH6.8, or 100% methanol at -20C. Permeabilise cells with0.5% Triron X-100). Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
