Specificity
|
M2III-5 reacts with an internal epitope of MRP2 (ABCC2), a 190-200 kDa transmembrane protein earlier known as the canalicular multi-organic anion transporter (cMOAT), which is absent in humans with the Dubin-Johnson syndrome, an autosomal recessive liver disorder characterized by chronic conjugated hyperbilirubinemia. MRP2 is a member of the MRP family of multidrug resistance related proteins. MRP2 is overexpressed in a subset of cisplatin-resistant cell lines. M2III-5 does not cross-react with the human MDR1 P-gp, MRP1, MRP3 or MRP5 gene products. Species Reactivity: Rat, mouse and human. Others not tested.

Application Notes
|
Immunocytochemistry: Use at a 1:20-1:50 dilution on acetone-fixed cytospin preparations. Immunohistochemistry: Use at a 1:20 dilution on acetone-fixed frozen sections, followed by incubation with rabbit anti-mouse IgG and a monoclonal mouse APAAP complex. M2III-5 can also be used on formaldehyde-fixed paraffin-embedded human tissues and tumors, after pretreatment with 0.01 M citric acid (pH 6.0) in distilled water at 100 °C for 3 x 5 minutes. After incubation with M2III-5 at a 1:20 dilution and washing, slides can be incubated with biotinylated rabbit anti-mouse IgG and streptavidin conjugated to horseradish peroxidase. Western blot: Use at a 1:20-1:50 dilution, followed by anti-mouse HRP. Flow cytometry: Optimal conditions still to be determined. The optimal dilution for a specific application should be determined by the researcher.
