| Specificity | Reacts with the 36 kDa Proliferating Cell Nuclear Antigen (PCNA), also known as cyclin or polymerase delta auxiliary protein. Elevated expression of PCNA/cyclin occurs in the nucleus during late G1 phase immediately before the onset of DNA synthesis. Expression becomes maximal during S-phase and declines during the G2 and M phases. PCNA reactivity is widely used as a prognostic marker for carcinogenic progression. Species Reactivity: Human, mouse, rat, insect and yeast. Others not tested. |

| Application Notes | Flow cytometry Immunohistology: For tissues fixed in methacarn, Bouin, or formalin and embedded in paraffin. Staining of formalin-fixed, paraffin-embedded tissues is enhanced by boiling the tissue sections in 10 mM citrate buffer, pH 6.0, for 10 min followed by cooling at RT for 20 min. Immunoprecipitation: Use 10 µL/mg protein lysate. Western blot: Use at a 1:400-1:800 dilution. The optimal dilution for a specific application should be determined by the researcher. |
