| Description | TCF4 (T cell factor 4, also known as TF7L2) serves as transcriptional activator affecting e.g.cell cycle and terminal cell differentiation. In the absence of Wnt signals, TCF4 interacts withTLE/Groucho corepressor, supressing the transcription of TCF4 target genes. Wnt signalingleads to stabilisation and accumulation of beta-catenin, which binds TCF4 N-terminus,whereas TCF4 C-terminal HMG-box domains interact with DNA. Cell death-associated Daxxprotein or plakoglobin belong to negative regulators of TCF4-mediated transcription, on theother hand, there is a positive feedback loop of beta-catenin-TCF4 pathway comprising thep300 coactivator. |
