Description: CaM Kinase IV (also known as CAM kinase-GR and CaMK IV) is a calcium/ calmodulin-dependent protein kinase belonging to a proposedcalcium-triggered signaling cascade. This kinasemay be involved in the transcriptional regulation ofmicrotubule dynamics. In vitro, CaMK IVphosphorylates CREB1, CREBBP, PRM2, MEF2A,MEF2D and STMN1/OP18. CaMK IV may also beinvolved in spermatogenesis and may play a role inthe consolidation/ retention of hippocampus-dependent long-term memory. CaMK IV must bephosphorylated to be maximally active and isphosphorylated by CAMKK1 or CAMKK2. Inaddition autophosphorylation of the N-terminus isrequired for full activation. Autophosphorylation ofSer-336 allows the kinase to switch to aCa(2+)/calmodulin-independent state. Most likelythe kinase is inactivated by the serine/ threonineprotein phosphatase 2A. CaMK IV is a monomerthat is located within the cytoplasm and nucleus andsubstantial localization occurs in certain neuronalnuclei. In spermatids CaMK IV is associated withchromatin and the nuclear matrix. CaMK IV is alsospecifically expressed in epithelial ovarian cancertissue.

Specificity: This antiserum is directed against human CaM Kinase IV protein. The product was delipidated,defibrinated followed by buffering and clarification. A BLAST analysis was used to suggest reactivity with this protein fromhuman, mouse, rat, dog, bovine and chimpanzee based on 100% homology for the immunogen sequence.

Western blot using antibodies-online's Anti-CaM Kinase IV antibody shows detection of a band ~52 kDa corresponding to CaM Kinase IV (arrowhead) in various preparations. Staining of rat brain lysate is shown in lane 1. Jurkat cell lysate staining is shown in lane 2. Specific reactivity is blocked in both lysates when antibody is preincubated with immunizing peptide (lanes 3 and 4 respectively). Approximately 35 ug of each lysate was separated by 4-20% SDS-PAGE and transferred onto nitrocellulose. CaM Kinase IV was similarly detected on lysates from mouse brain (not shown). After blocking the membrane was probed with the primary antibody diluted to 1:1,000 for 2h at room temperature followed by washes and reaction with a 1:10,000 dilution of IRDye
